 2016年,人類細胞圖譜計劃(HumanCell Atlas Project)被提出:根據*的分子信息(如基因表達)對所有人類細胞種類進行定義,并將這些信息與傳統的細胞學表述(如位置和形態)相關聯,來了解人體中的所有細胞的類型、每個細胞的空間位置、每個細胞在不同生命時刻的狀態以及不同個體細胞圖譜的疊加,并根據不同健康狀態、基因型、生活方式和外界環境加以注釋。
 
       而傳統的單細胞測序技術只是對單個細胞進行轉錄組學的研究,很難實現大通量單細胞轉錄組測序;近年來,單細胞測序技術獲得快速發展,尤其是基于微液滴技術的大規模單細胞分選平臺的出現,使我們能夠以較低的成本對某一器官或者組織構建其細胞表達圖譜,了解各個器官或者組織細胞復雜的基因網絡在單個細胞中的運作機制,并了解各個細胞中這些網絡的差異,以及zui終各種細胞群體如何協同工作。

       Chromium™ Single Cell 3' Solution是一種在單個細胞水平對mRNA進行高通量測序的新技術,為單細胞轉錄組研究提供更具擴展性的平臺。其基于10X Genomics平臺,一次性能夠分離500~10000個單細胞,并能夠將分離的單個細胞中的微量mRNA通過高效擴增后再進行高通量測序,有效地解決了組織樣本無法破解的細胞異質性難題,有助于發現新的細胞類型;并基于大量的單細胞基因表達數據,可進行細胞表達特征聚類、亞群表達特征、標志物篩選等方面的分析,是一種高效的細胞基因表達水平的檢測技術,在生殖、免疫反應、干細胞發育與分化、腫瘤異質性、神經系統研究以及腦發育等研究領域中有著廣泛的應用。
